What is enzyme immobilization?
Enzymes are nature's perfect catalysts. So why are they so hard to use at industrial scale — and what does immobilization have to do with it?
18 June 2026Enzymes are remarkable. They catalyse chemical reactions with a precision that no synthetic chemist has managed to replicate: high selectivity, mild conditions, no toxic by-products. In a world increasingly concerned with the environmental footprint of manufacturing, they look like an obvious solution.
And yet, if you talk to anyone who has tried to use an enzyme in an industrial process, you’ll hear the same story: enzymes are expensive, they’re sensitive, and, the real pain, they’re single-use. Produce, purify, use, repeat.
Run a reaction, separate your product, and the enzyme ends up in the waste stream. At laboratory scale, that’s annoying. At industrial scale, it’s often what tips the economics from viable to impossible.
What immobilization actually involves
In practice, there are many ways to attach an enzyme to a surface. You can adsorb it (weak, reversible, often leaches over time), covalently bond it (stronger, but risks deforming the protein), entrap it in a gel (protects it, but limits diffusion), or cross-link enzyme aggregates into stable particles.
Each method has trade-offs, and the right choice depends on the specific enzyme, the reaction conditions, the solvent, the pH. There’s no universal answer.
What makes immobilization genuinely difficult is that the enzyme is not a passive molecule. It’s a folded, three-dimensional structure, and attaching it in the wrong place or in a way that distorts its shape can destroy it and make it useless. You might end up with an enzyme that is firmly stuck to your support and completely inactive.
This is why choosing where and how to immobilize an enzyme is not just a chemistry problem. It’s also a structural biology problem. The active site needs to remain accessible. Flexible regions that are important for catalysis need to stay flexible. The surface chemistry of the carrier needs to be compatible with the enzyme’s surface. And so many more parameters.
Why it matters for green chemistry
When immobilization works, the gains are significant. Enzyme reuse directly reduces cost. Immobilized enzymes also tend to be more stable — partially because the support can protect them from aggregating or unfolding. This opens up reaction conditions (higher temperatures, harsher solvents) that a free enzyme wouldn’t tolerate.
The bigger unlock is continuous flow. Pack a column with immobilized enzyme, pump your substrate through, and collect product at the other end. No batch-to-batch variation, no downtime between runs, much easier to scale. Flow biocatalysis has become one of the central strategies for sustainable pharmaceutical manufacturing, and immobilization is what makes it work.
The bioinformatics angle
For most of my research career, the question I kept coming back to was: can we be smarter about how we choose immobilization conditions? Instead of exhaustive experimental screening — trying every combination of support and coupling chemistry until something works — can we use protein structure data to make better predictions upfront?
That question led to CapiPy, a bioinformatics tool for analysing immobilization candidates, and eventually to inseit, which is building an AI-powered platform to take this further.
The premise is that the enzyme already contains the answer — in its structure, its surface chemistry, the location of its active site. We just need better tools to read it.
